Exploiting Pan Influenza A and Pan Influenza B Pseudotype Libraries for Efficient Vaccine Antigen Selection

چکیده

We developed an influenza hemagglutinin (HA) pseudotype library encompassing Influenza A subtypes HA1-18 and B (both lineages) to be employed in microneutralization (pMN) assays. The pMN is highly sensitive specific for detecting virus-specific neutralizing antibodies against viruses can used assess antibody functionality vitro. Here we show the production of these viral HA pseudotypes their employment as substitutes wildtype neutralization demonstrate utility serum responses vaccination with ability evaluate cross-subtype elicited by vaccinating antigens. Our findings may inform further preclinical studies involving immunization dosing regimens mice help creation selection better antigens vaccine design. These harnessed meet strategic objectives that contribute strengthening global surveillance, expansion seasonal prevention control policies, pandemic preparedness response.

Pan-Influenza A Protection by Prime–Boost Vaccination with Cold-Adapted Live-Attenuated Influenza Vaccine in a Mouse Model

Influenza virus infections continually pose a major public health threat with seasonal epidemics and sporadic pandemics worldwide. While currently licensed influenza vaccines provide only strain-specific protection, antigenic drift and shift occasionally render the viruses resistant to the host immune responses, which highlight the need for a vaccine that provides broad protection against multi...

Influenza vaccine strain selection strategies

Currently, the influenza vaccine is updated only when there is at least a 4-fold difference [measured by the hemagglutination inhibition (HI) assay] between the current vaccine strain and an expected epidemic strain. It is not clear if this is the optimal strategy, especially for individuals vaccinated every year. Using a computer model, we determined the vaccine efficacy resulting from 12 diff...

Characterization of Influenza Vaccine Immunogenicity Using Influenza Antigen Microarrays

BACKGROUND Existing methods to measure influenza vaccine immunogenicity prohibit detailed analysis of epitope determinants recognized by immunoglobulins. The development of highly multiplex proteomics platforms capable of capturing a high level of antibody binding information will enable researchers and clinicians to generate rapid and meaningful readouts of influenza-specific antibody reactivi...

Expression of HA1 antigen of H5N1 influenza virus as a potent candidate for vaccine in bacterial system

The impending influenza virus pandemic requires global vaccination to prevent large-scale mortality and morbidity, but traditional influenza virus vaccine production is too slow for rapid responses. In this study, bacterial system has been developed for expression and purification of properly folded HA1 antigen as a rapid response to emerging pandemic strains. Here, a recombinant H5N1 (A/Indone...
